I have an Alesis QSR, and I had the same problem. First, you need the instrument definitions for your particular synth installed into Cakewalk, and secondly, you need to make sure that Cakewalk is set up to receive and send signals to your synth.
My particular set up is that I use the Alesis serial port driver, and the serial port cable that I bought from a music store. The serial port cable is connected to the serial port on both my QSR and my PC. In my sound configuration in Windows, the serial port is setup for MIDI. When I run Cakewalk, I have Cakewalk setup to send and receive signals from the serial port, and I have the Alesis set up for serial I/O.

I'm not sure what you're using, so it's difficult for me to give you any specific advice.

I might add that you can access the sound banks in an Alesis keyboard if you have the correct instrument definitions installed.
